About Lion's mane

An edible white toothed fungus that grows on hardwood, eaten as food in East Asia and sold as an extract for cognitive claims. Its studied compounds are hericenones from the fruiting body and erinacines from the mycelium.

Lion's mane is the mushroom that nootropic marketing has adopted most enthusiastically, on the basis of a genuinely interesting mechanism and a genuinely thin clinical record. In cell culture, hericenones and erinacines stimulate nerve growth factor synthesis. In people, the anchor study is a Japanese trial in about thirty older adults with mild cognitive impairment, taking 3 g of powder daily for sixteen weeks, in which cognitive scores improved during supplementation and returned toward baseline after it stopped. That is a small, single-centre result and subsequent trials have been few and mixed. A structural detail matters commercially: hericenones occur in the fruiting body and erinacines in the mycelium, so fruiting-body and mycelium products do not contain the same compounds, and neither has been shown superior in a human trial.

One small Japanese trial in older adults with mild cognitive impairment reported improved cognitive scores during sixteen weeks of supplementation, fading after it stopped. A few short trials in healthy adults have reported small changes in processing speed. The total human evidence is a few hundred participants and does not yet support a cognitive claim.

What may be going on: Hericenones and erinacines stimulate nerve growth factor synthesis in cultured cells. Whether either compound crosses into human brain tissue at supplement doses has not been shown.

  • Allergy

    Respiratory and skin reactions reported

    Case reports describe acute breathing difficulty and skin eruptions after lion's mane ingestion, including one report of respiratory failure. Mushroom allergy is uncommon but real, and anyone with known mould or fungal sensitivity should approach cautiously.

  • Worth knowing

    Fruiting body and mycelium are different products

    Hericenones are found in the fruiting body, erinacines in the mycelium. A capsule of grain-grown mycelium and a capsule of fruiting-body extract contain different compounds, and the human trial evidence used dried fruiting body. Neither the substitution nor its consequences have been tested in people.

    Antiplatelet and glucose-lowering effects have been described in animal work. Human interaction data are absent, which is a reason to mention it to a prescriber rather than a reason for reassurance.

    The cognitive case rests largely on one trial of roughly thirty people. Products marketed for focus, memory and nerve regeneration are extrapolating a long way from that, and long-term safety at supplement doses has not been studied.
