Muscle protein synthesis
Casein raises muscle protein synthesis and, taken before sleep, raises it overnight — a period otherwise spent in net protein breakdown. Training trials adding pre-sleep casein report greater gains in strength and muscle size, though whether the timing or the additional protein is responsible is not fully separated.
What may be going on: Gastric clotting slows emptying, producing a prolonged low-amplitude rise in plasma amino acids rather than a sharp peak.
Moderate evidence. Several human studies point the same way, with real caveats.
