About Branched-chain amino acids (BCAAs)

The three branched-chain essential amino acids — leucine, isoleucine and valine — sold as a free-form powder, usually in a 2:1:1 ratio. They are already present in any complete protein at roughly 20% of its amino acid content.

BCAAs are the clearest case in this catalogue of a supplement whose mechanism is real and whose product is redundant. Leucine does trigger muscle protein synthesis. But building muscle protein requires all the amino acids, and supplying three of them without the other six essentials produces a smaller synthetic response than whole protein — measured directly in a 2017 study where BCAAs raised muscle protein synthesis by 22% while whey in the same design raised it far more. Reviewers have concluded that BCAA supplementation cannot support increased muscle protein synthesis in humans on its own. Where total protein intake is adequate, which is the situation of nearly everyone buying them, the expected effect on muscle growth is nil.

What is in a retail bottle is frequently not what was studied, and the amount that suits one person is not general advice.

What each association actually rests on, and the papers behind it. A rating describes the state of the research, not how promising the supplement sounds. How we rate evidence.

Meta-analyses report small reductions in delayed-onset muscle soreness and in creatine kinase after damaging exercise with BCAAs. The trials are small, often use untrained participants and low-protein control conditions, and the effect on actual recovery of function is inconsistent.

The central fatigue hypothesis — that BCAAs reduce brain tryptophan uptake and delay perceived fatigue — has not produced consistent performance benefits in trials. There is not enough evidence to support a performance claim.

Where dietary protein is adequate, BCAA supplementation has not been shown to increase muscle mass or strength beyond what the protein already provides. Direct measurement shows BCAAs alone raise muscle protein synthesis considerably less than an equivalent amount of complete protein, because the other essential amino acids required to build protein are missing. This is a largely null finding, not an absence of research.

What may be going on: Leucine activates mTORC1 and initiates translation, but translation requires the full complement of amino acids. Supplying leucine without them raises the signal without the substrate.

    Redundant when protein intake is adequate

    A 25 g serving of whey already contains roughly 5–6 g of BCAAs alongside every other amino acid needed to build muscle protein. Buying BCAAs on top of adequate protein is paying separately for a subset of something already consumed.

    Competes with other amino acids for transport

    Large free-form BCAA doses compete with tryptophan, tyrosine and other large neutral amino acids at the blood-brain barrier transporter, and can lower their central availability. The clinical meaning in healthy people is unclear, but it is not a neutral intervention.

    Metabolic associations

    Elevated circulating BCAAs are consistently associated with insulin resistance in observational metabolic research. Whether supplementation contributes to that or merely reflects it is unresolved, but it is a reason not to assume large doses are inert.

    Maple syrup urine disease

    BCAA supplements are contraindicated in maple syrup urine disease, an inherited disorder of branched-chain amino acid metabolism, and require care in advanced liver disease outside a supervised protocol.
